dc.description.abstractAntioksidan adalah senyawa yang mampu menangkal atau meredam efek negatif oksidan dalam tubuh, bekerja dengan cara mendonorkan satu elektronnya kepada senyawa yang bersifat oksidan sehingga aktivitas senyawa oksidan tersebut dapat dihambat. Tujuan penelitian ini adalah untuk mengetahui karakteristik simplisia, skrining fitokimia, ekstraksi secara maserasi serta aktivitas antioksidan dengan metode DPPH, penentuan kandungan fenolat dan flavonoid total dari ekstrak etanol herba poguntano. Pemeriksaan karakteristik simplisia meliputi pemeriksaan makroskopik, mikroskopik, penetapan kadar air, penetapan kadar sari yang larut dalam air, penetapan kadar sari yang larut dalam etanol, penetapan kadar abu total dan penetapan kadar abu yang tidak larut asam. Skrining fitokimia terhadap serbuk simplisia meliputi pemeriksaan golongan senyawa alkaloid, flavonoid, glikosida, glikosida antrakuinon, tanin, saponin dan steroid/triterpenoid. Ekstrak diperoleh secara maserasi dengan pelarut etanol 96%, lalu dipekatkan dengan alat rotary evaporator hingga didapatkan ekstrak kental. Ekstrak diuji dengan metode pemerangkapan radikal bebas DPPH (1,1-diphenyl-2-picrylhydrazyl) serta mengukur kandungan fenolat total dan kandungan flavonoid total dengan mengukur absorbansi menggunakan spektrofotometri uv-vis yang telah didiamkan selama 60, 90 dan 40 menit pada panjang gelombang 516, 775 dan 432 nm. Hasil karakteristik simplisia herba poguntano diperoleh kadar air 7,99%, kadar sari larut air 18,51%, kadar sari larut dalam etanol 7,36%, kadar abu total 9,58% dan kadar abu yang tidak larut dalam asam 0,82%. Hasil skrining fitokimia serbuk simplisia mengandung senyawa alkaloid, flavonoid, glikosida, glikosida antrakuinon, tanin, saponin dan steroid/triterpenoid. Hasil pengukuran aktivitas antioksidan dengan metode pemerangkapan radikal bebas DPPH, ekstrak etanol herba poguntano menunjukkan kekuatan antioksidan dalam kategori sangat kuat dengan nilai IC50 21,79 ppm dan kuersetin juga menunjukkan kekuatan antioksidan dalam kategori sangat kuat dengan nilai IC50 4,95 ppm. Hasil pengukuran kandungan fenolat menunjukkan angka 66,29 mg GAE/g ekstrak serta hasil pengukuran kandungan flavonoid total menunjukkan angka 19,26 mg QE/g ekstrak.en_US
dc.description.abstractAntioxidants are compounds that can counteract or mitigate the negative effects of oxidants in the body, works by donating an electron to compounds that are oxidant so that the oxidant compound activity can be inhibited. The purpose of this study was to determine the characteristics, phytochemical screening, maseration extraction and antioxidant activity by the method of DPPH as well as knowing the content of total phenolics and flavonoids from the ethanol extract of the herb poguntano. Simplex characterization including macroscopic, microscopic, level of water content, level of water-soluble extract, level of ethanol-soluble extract, level of total ash and level of ash not soluble in acid. Phytochemical screening of the simplex powder including determination alkaloids, flavonoids, glycosides, antracuinon glycosides, saponin, tannins, and steroids/triterpenoids. The extract obtained by maceration with 96% ethanol, and then concentrated by means of a rotary evaporator to obtain a thick extract. Extract were tested by the method of trapping free radicals DPPH (1,1-diphenyl-2-picrylhydrazyl) as well as measure the concentration of total phenolics and flavonoids by measuring the absorbance using spectrophotoeter uv-visible spectrophotometry which had been allowed to stand for 60, 90 and 40 minutes at a wavelength of 516, 775 and 432 nm. The results of the simplex characterization obtained level of water content 7.99%, level of water soluble extract 18.51%, level of ethanol soluble extract 7.36%, level of total ash 9.58% and level of ash not soluble in acid 0.82%. Phytochemical screening results simplex powder containing compounds alkaloids, flavonoids, glycosides, antracuinon glycosides, saponin, tannins and steroids/triterpenoids. Results of measuring the antioxidant activity with DPPH free radical trapping method, the ethanol extract of the herb poguntano shows the antioxidant power in the strong category with IC50 values 21.79 ppm and quercetin shows the antioxidant power in very strong category with IC50 values of 4.95 ppm. Results by measuring total phenolics content shows 66.29 mg GAE/g extract and results by measuring total flavonoid content shows 19.26 mg QE/g extract.en_US
