dc.description.abstractLatar Belakang : Daun biwa (Eriobotrya japonica (Thunb.) Lindl) telah digunakan untuk mengobati penyakit saluran napas. Namun, mekanisme aksinya untuk mengobati penyakit saluran napas belum banyak dilaporkan. Tujuan : Untuk mengetahui apakah ekstrak etanol daun biwa (EEDB) memiliki efek relaksasi terhadap organ trakea marmut terisolasi yang diinduksi histamin melalui jalur nitric oxide dan prostaglandin. Metode : Meliputi skrining fitokimia dan uji efek relaksasi menggunakan trakea marmut. Parameter yang diukur adalah kontraktilitas otot polos trakea marmut terisolasi. Trakea marmut diekuilibrasi selama 45 menit dengan larutan Kreb’s suhu 37oC yang diaerasi gas karbogen. Pengujian efek relaksasi dilakukan setelah trakea marmut dikontraksi dengan histamin EC80 (4,62×10-4M), kemudian diberikan konsentrasi kumulatif EEDB 1-8 mg/ml. Pengujian mekanisme efek relaksasi EEDB dilakukan dengan menginkubasi trakea menggunakan L-NAME 10-4M dan aspirin 10-4M selama 20 menit kemudian dikontraksi dengan histamin EC80, selanjutnya diberikan konsentrasi kumulatif EEDB 1-8 mg/ml. Hasil : Hasil penelitian menunjukkan bahwa EEDB mampu merelaksasi otot polos trakea yang diinduksi histamin. Terdapat pengaruh antara peningkatan konsentrasi dengan efek relaksasi, relaksasi maksimal terjadi pada konsentrasi 8 mg/ml dengan relaksasi mencapai 40%. Terdapat perbedaan relaksasi antara trakea yang diinkubasi dengan L-NAME, aspirin dan tanpa inkubasi (p=0.014). Kesimpulan : EEDB memiliki efek relaksasi terhadap otot polos trakea marmut terisolasi yang diinduksi histamin. Mekanisme efek relaksasi EEDB dimediasikan melalui prostaglandin dan jalur nitric oxide.en_US
dc.description.abstractBackground: Biwa leaves (Eriobotrya japonica (Thunb.) Lindl) have been used to treat respiratory disease, but the mechanism of action of these leaves to treat respiratory disease has not been widely reported. Objective: To determine whether the ethanol extract of the Eriobotrya japonica leaf (EEEJL) has a relaxing effect on isolated marmot trachea induced with histamine through nitric oxide and prostaglandin pathway. Method: Including phytochemical screening and the effect of relaxation was tested using the marmot trachea. The parameters measured in this study were contractility of smooth muscle isolated from the marmot trachea. The marmot trachea was equilibrated for 45 minutes in Kreb's solution was obtained at 37°C aerated with carbogen gas. The relaxation effect testing was done after the marmot trachea have been contracted with histamine (EC80: 4.62 × 10-4M), then given the cumulative concentration of EEEJL 1-8 mg/ml. The mechanism of the EEEJL relaxation effect testing was done by incubating the trachea with L-NAME 10-4 M and aspirin 10-4M for 20 minutes then contracted with histamine, then given a cumulative concentration of EEEJL 1-8 mg/ml. Result: The results showed that the EEEJL was able to relax histamine-induced tracheal smooth muscle. There is an effect between increased concentration on the tracheal relaxation, the maximum relaxation occurs at the concentration of 8 mg/ml with relaxation reaching 40%. There is a significant differences between L-NAME and aspirin incubated trachea and non-incubated one (p= 0.014). Conclusion: EEEJL has a relaxation effect on isolated tracheal smooth muscle of marmot induced by histamine. The mechanism of the EEEJL relaxation effect is mediated through prostaglandin and nitric oxide pathway.en_US
