dc.description.abstractFifteen isolates of Metarhizium anisopliae isolated from 15 oil palm plantation sites in North Sumatra were examined for their morphological and molecular diversity as well as their virulence against O. rhinoceros larvae. On the basis of morphological characteristics, all isolates were identified as M. anisopliae. The results of genetic variability analysis of M. anisopliae isolates by PCR random amplified polymorphism DNA (RAPD) technique formed 3 groups.The result of sequencing with the ITS primer showed that all isolates of M. anisopliae isolated from soil had a homology of 85-99% with M. anisopliae reference strains from genbank database NCBI. The results of M. anisopliae virulence test against O. rhinoceros larvae, isolate from O. rhinoceros larvae against 3rd instar larvae (M0L3) was the highest mortility percentage (20%), followed by isolate from O. rhinoceros larvae against 2nd instar larvae (M0L2), isolate from Namotualang against 3rd instar larvae (M14L3), isolate from Bandar Rejo against 2nd instar larvae (M2L2), isolate from Kampung Baru 2nd instar larvae (M1L2), isolate from Bandar Selamat against 2nd instar larvae (M3L2), isolate from Bandar Tonga against 2nd instar larvae (M8L2), isolate from Bandar Tonga against 3rd instar larvae (M8L3), isolate from Lias Baru against 2nd instar larvae (M9L2) and isolate from Perdamaian against 2nd instar larvae (M10L2) (13.33%, 10%, 6.67%, 3.33%, 3.33%, 3.33%, 3.33%, 3.33%, 3.33%, and 3.33% respectively). Virulence was categorized as low because it was under 30%. Other isolates could not kill O. rhinoceros larvae.en_US
dc.description.abstractLima belas isolat Metarhizium anisopliae yang diisolasi dari 15 lokasi tanah pertanaman kelapa sawit di Sumatera Utara diteliti keragaman morfologi dan molekulernya dan juga virulensinya terhadap larva O. rhinoceros. Secara morfologi semua isolat diidentifikasi sebagai M. anisopliae. Hasil analisis keragaman genetik isolat-isolat M. anisopliae dengan teknik PCR random amplified polymorphism DNA (RAPD) membentuk 3 kelompok. Hasil sekuensing dengan primer ITS menunjukkan semua isolat M. anisopliae yang diisolasi dari tanah memiliki homologi 85-99% dengan strain M. anisopliae yang dirujuk berdasarkan database genbank NCBI. Hasil uji virulensi M. anisopliae terhadap larva O. rhinoceros tertinggi pada perlakuan isolat asal larva O. rhinoceros terhadap larva instar 3 (M0L3) sebanyak 20%, diikuti berturut-turut isolat asal larva O. rhinoceros terhadap larva instar 2 (M0L2), isolat asal Namotualang terhadap larva instar 3 (M14L3), isolat asal Bandar Rejo terhadap larva instar 2 (M2L2), isolat asal Kampung Baru terhadap larva instar 2 (M1L2), isolat asal Bandar Selamat terhadap larva instar 2 (M3L2), isolat asal Bandar Tonga terhadap larva instar 2 (M8L2), isolat asal Bandar Tonga terhadap larva instar 3 (M8L3), isolat asal Lias Baru terhadap larva instar 2 (M9L2) dan isolat asal Perdamaian terhadap larva instar 2 (M10L2) (13.33%, 10%, 6.67%, 3.33%, 3.33%, 3.33%, 3.33%, 3.33%, 3.33%, dan 3.33% secara berurutan). Virulensi dikategorikan rendah karena berada dibawah 30%. Sedangkan untuk isolat perlakuan lainnya tidak bisa membunuh larva O. rhinoceros.en_US
