dc.description.abstractDiabetes mellitus (DM) is a chronic disease or metabolic disorder which can be identified by high blood glucose levels with metabolic disorders of carbohydrates, lipids and proteins as a result of insulin function insufficiency. The prevention of type II DM can be done by inhibiting α-glucosidase so the absorption of glucose is inhibited and the level of postprandial glucose is decrease. This study aims to know the activity and the mechanism of α-glucosidase and kinetics inhibition by the ethanol extract of Saurauia leaves. The dried powder of Saurauia leaves was extracted with ethanol 96%. The inhibitory activity and kinetic of enzyme test were measured by spectrophotometry using microplate reader at wavelength of 405 nm. Kinetic test was done to determine the inhibitory type of enzyme using inhibitor and without inhibitor to get Lineweaver-Burk plot. Statistied analysis was done by Independent- sample T test. The result showed that the ethanol extract of Saurauia leaves could inhibit α-glucosidase activity with the mean of IC50 75.64 ± 0.56 μg/mL which the result was weaker than acarbose with the mean of IC50 131.57± 0.83 μg/mL showing, there was a significant different which was 0.00 (p<0.05). Result of kinetics test obtained from this experiment is a competitive inhibitory type. It could be concluded of this experiment that ethanol extract of Saurauia leaves have α-glucosidase inhibitory activity the flavonoid compounds are isoflavons that are suspected as α-glucosidase inhibitor and have a competitive mechanism of the enzyme inhibition.en_US
dc.description.abstractDiabetes mellitus (DM) merupakan suatu penyakit atau gangguan metabolisme kronis yang ditandai dengan tingginya kadar glukosa darah disertai dengan gangguan metabolisme karbohidrat, lipid, dan protein sebagai akibat insufisiensi insulin. Salah satu pendekatan untuk mencegah diabetes mellitus tipe II adalah dengan cara menghambat α-glukosidase sehingga absorpsi glukosa tertunda dan kadar glukosa postprandial menurun. Tujuan dari penelitian untuk mengetahui aktivitas dan mekanisme penghambatan enzim α-glukosidase oleh ekstrak etanol daun pirdot (EEDP). Serbuk simplisia daun pirdot diekstraksi dengan menggunakan pelarut etanol 96 %. Pengujian aktivitas penghambatan enzim dan uji kinetika enzim dilakukan dengan metode spektrofotometri menggunakan microplate reader pada panjang gelombang 405 nm. Uji kinetika dilakukan untuk menentukan tipe penghambatan enzim dengan menggunakan inhibitor maupun tanpa inhibitor untuk mendapatkan plot Lineweaver-Burk. Uji statistik dilakukan dengan analisis Independent- sample T test. Hasil penelitian diperoleh EEDP dapat menghambat aktivitas enzim α-glukosidase dengan nilai rata-rata IC50 75,64 ± 0,56 μg/mL hasilnya lebih rendah dari nilai rata-rata IC50 akarbose yaitu 131,57 ± 0,83 μg/mL dengan menunjukkan perbedaan yang signifikan 0,00 (p<0,05). Hasil uji kinetika diperoleh jenis penghambatan kompetitif. Kesimpulan dari penelitian ini bahwa ekstrak etanol daun pirdot memiliki aktivitas penghambatan enzim α-glukosidase senyawa flavonoid berupa isoflavon yang berperan sebagai penghambat enzim α-glukosidase dan memiliki mekanisme inhibisi enzim yang kompetitif.en_US
