    Abstract
    The isolation, characterization and measurement of antibacterial activity of phenolic compound from E. tirucalli L has been conducted. Isolation process of phenolic compound is started by maceration with methanol and continued by separation process with chromatographic column. Antibacterial activity of the total phenolic extract is determined by agar difussion method. The phytochemical screening showed the methanolic crude extract of E. tirucalli contained phenolic compound. The total phenolic extract has antibacterial activity against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, dan Bacillus polymyxa with inhibition zone 10, 11, 14 and 15 mm. The maximum of inhibition zone is showed at concentration of 100 ppm against E.coli. The elucidation structure analysis of FT-IR, UV-Vis,1H-NMR,13C-NMR, and LC-MS showed the isolated compound is a derivative of gallic acid.
     
    Telah dilakukan isolasi dan karakterisasi senyawa fenolik dari tanaman patah tulang (E. tirucalli L), serta uji aktivitas antibakteri. Proses isolasi senyawa fenolik diawali dengan maserasi menggunakan pelarut metanol dan dilanjutkan dengan proses pemisahan menggunakan kromatografi kolom. Aktivitas antibakteri dari ekstrak fenolik total ditentukan menggunakan metode difusi agar. Hasil uji skrining fitokimia menunjukkan bahwa ekstrak kasar metanol E. tirucalli mengandung senyawa golongan fenolik. Ekstrak fenolik total memiliki aktivitas antibakteri terhadap Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, dan Bacillus polymyxa dengan zona hambat 10, 11, 14 dan 15 mm. Penghambatan maksimum ditunjukkan pada konsentrasi 100 ppm terhadap E. coli. Elusidasi struktur senyawa hasil isolasi berdasarkan analisis FT-IR, UV-Vis, 1H-NMR, 13C-NMR, dan LC-MS menunjukkan bahwa senyawa hasil isolasi dari ekstrak metanol E. tirucalli L merupakan turunan asam galat.
