Wounds are damage to body tissues, caused by mechanical, chemical, thermal, and/or physical factors. Untreated wounds can lead to infectious diseases such as tetanus. One of the treatment options to treat infections is by utilizing plants or plants that are known to have antimicrobial/antibacterial properties. Iler leaves contain several compounds such as flavonoids, glycosides, tannins, saponins, and steroids or triterpenoids. Because it has antimicrobial properties, this substance can accelerate wound healing. Iler leaf extract is made in gel preparations because gel preparations are easy to use and easily penetrate the skin. To formulate ethanol extract of iler leaves (Coleus scutellarioides (L.) Benth) in the form of a stable gel preparation and to determine the effectiveness of ethanol extract gel preparations of iler leaves as a healer of excision incision wounds in rats induced by Staphylococcus aureus.
The stages of the research carried out were the manufacture of ethanol extract of iler leaves, examination of characteristics, phytochemical screening, antibacterial testing of Staphylococcus aureus, preparation of ethanol extract gel of iler leaves, evaluation of gel preparations, and stability of preparations, manufacture of excision wounds of rats induced by Staphylococcus aureus, as well as the treatment and observation of the wounds of test animals. The rats were divided into 5 groups. The wound was smeared with a gel of ethanol extract of iler leaves twice a day for 14 days with different extract concentrations of 5%, 7.5%, and 10%, with Bioplacenton® as the positive control and the gel base as the negative control. The results of this study showed that ethanol extract of iler leaves (Coleus scutellarioides (L.) Benth) could be formulated in a stable gel preparation for 3 months. Ethanol extract gel preparation of iler leaves (Coleus scutellarioides (L.) Benth) has the effect on healing excision cut wounds in rats induced by Staphylococcus aureus. The most effective concentration, namely the ethanol extract gel of iler leaves with a concentration of 10%, has a percentage of wound healing area of 93.23±0.87%. Extract ethanol iler leaves can be formulated in a stable gel preparation for 3 months and has an effect on the healing of excision cut wounds in rats induced by Staphylococcus aureus.
