Test of Total Fenol Content of Hydroglycolic Extract of Puguntano Leaves (Picria fel-terrae Lour.) and Antioxidant Activity Test by DPPH Method
Abstract
Background: There are several factors that influence the extraction process. This will affect the acquisition of the level of an active substance compound, one of which is the concentration of the extracting solvent and the ratio of the sample and the solvent used.
Objective: This study aims to determine the total phenol content and antioxidant activity of hydroglycolic extract of puguntano leaves.
Methods: This study was conducted experimentally including sample preparation, preparation of hydroglycolic extract of puguntano leaves, simplicia characterization, phytochemical screening, determination of total phenol content and antioxidant activity testing.
Results: The results of total phenol content of hydroglycolic extract of puguntano leaves are best at a solvent concentration of 80% with a ratio (1: 10) has a value of
25,03 ± 1,33 mg GAE / g extract. The results of antioxidant activity of vitamin C solution (positive control) are with an IC50 value of 3,09 µg/ml, while the best antioxidant activity of puguntano leaf hydroglycolic extract is at a solvent concentration of 80% with a ratio (1:10), namely with an IC50 value of 189,79 ± 1,30 with a weak category.
Conclusion: The results showed that the hydroglycolic extract of puguntano leaves contained total phenol content with a value of 25,03 ± 1,33 mg GAE/g extract and antioxidant activity with a IC50 value of 189,79 ± 1,30 (weak).
