dc.description.abstract | Irrigation is very important in root canal treatment. Sodium hypochlorite (NaOCl) is the gold standard irrigation material, but has the disadvantage of being toxic if extruded into periapical tissues. As an alternative, quantum dots (QDs) lerak extract containing active compounds of saponins, alkaloids, tannins, flavonoids and polyphenols, potentially have antibacterial activity. Although QDs lerak extract can be developed as a root canal irrigation material, it is not yet known how they affect cytotoxicity against 3T3 fibroblast cells. Therefore, the purpose of this study was to determine the difference in cytotoxicity of QDs lerak extract on the growth of 3T3 fibroblast cells and the difference with 2.5% NaOCl. This type of research is laboratory experimental research with post test only control group design. There were 8 groups consisting of 100%, 50%, 25%, 12.5%, 6.25%, 3.125%, 1.625% QDs lerak extract solution, and 2.5% NaOCl. Cell cytotoxicity testing was carried out by observing the viability of 3T3 fibroblast cells that had been given the test solution and then given MTT solution by reading the absorbance of the Elisa reader after a 24-hour incubation period. Data obtained were subjected to One way ANOVA statistical test and followed by Games Howell test. One-way ANOVA test results showed a significant difference in 3T3 fibroblast cell viability between all treatment groups (p<0.05). The QDs lerak extract group with a concentration of 6.25% produced the highest cell viability, which indicates the lowest potential cytotoxicity compared to other treatment groups. On the other hand, the 2.5% NaOCl group showed a very high level of toxicity compared to the QDs lerak extract group. | en_US |
