dc.description.abstractHigh sugar diet is often associated with increased blood sugar (hyperglycemia). Hyperglycemia that occurs continuously can trigger an imbalance between increased ROS production through mitochondrial dysfunction and reduced antioxidant defenses. An increase in ROS causes DNA damage and then causes oxidative stress. The antioxidant defense system consists of endogenous antioxidants, one of which can be assessed from the expression of the sod1 and sod2 genes. Pirdot leaves (saurauia vulcani korth.) contain bioactive compounds such as flavonoids, penolics, saponins, tannins and triterpenoids. The methods of the research was a true experimental posttest control group study using 10 drosophila melanogaster animals divided into 5 groups, namely Kn: group without treatment (D.melanogaster given normal food); Kp: group fed a high sugar diet; P1: group given a high sugar diet and 0.01% pirdot extract, P2: group given a high sugar diet and 0.1% pirdot extract and P3: group given a high sugar diet and 1% pirdot extract. Determination of gene expression using real-time PCR (RT-qPCR. The results showed that there were differences in the hemolymph levels of Drosophila melanogaster larvae between the normal diet group and the high sugar diet group. treatment group. In molecular tests, the results showed that pirdot leaf extract could increase the expression levels of the sod1 and sod2 genes, which began to increase at concentrations of 0.1% and 1%. In the locomotor test, it was seen that the pirdot leaf extract had a significant effect on locomotory in both male and female groups (p<0.0001). Conclusion: Pirdot leaf ethanol extract has the potential as an exogenous antioxidant in d.melanogaster hyperglycemia models by increasing the expression levels of sod1 and sod2 (the effect begins to be seen at concentrations of 0.1 and 1%) and can also improve locomotor disorders so that it can ward off ROS due to a high sugar diet.en_US
