dc.description.abstract | Background : In Southeast Asia, especially Indonesia, mobe leaves are used as medicine because they contain secondary metabolite components like flavonoids, tannins, saponins, and glycosides. Flavonoids in mobe leaves are the source of quercetin and apigenin.
Objective: The purpose of the study was to compare the content of quecetin and apigenin from four different extraction methods, as follow: maceration, percolation, reflux, soxhlet using UHPLC method.
Method: This research includes making mobe leaves ethanol extract using the maceration, percolation, reflux, soxhlet methods and characterization of simplicial and extract which includes determination of water content, water soluble essence content, ethanol soluble essence content, total ash content, acid insoluble ash content, phytochemical sreening, qualitative test measurement with UHPLC and method validation.
Results: The results of the ethanol extract of mobe leaves using the maceration extraction method obtained a yield of. 9%, with the percolation extraction method the yield was 19.7%, with the reflux extraction method the yield was 7.26%, with the Soxhlet extraction method the yield was 13.86%. The results of the UHPLC research showed that the chromatogram of mobe extract showed that there were apigenin and quercetin compounds. The apigenin content values of maceration, percolation, reflux, Soxhlet extracts were 0.0233±0.0001365; 0.1429±0.004029; 0.0117±0.0007283; 0.0316±0.0005384. The highest apigenin levels were obtained by the percolation extraction method. The quercetin content values of maceration, percolation, reflux, Soxhlet extracts were 0.1717±0.001631; 0.1252±0.001655; 0.0731±0.001052; 0.0194 ±0.002506. The highest quercetin levels were obtained by the maceration extraction method. Validation test results obtained % recovery of apigenin 104.62%, quercetin 108.76%; LOD apigenin 0.0013644 mg/g dan LOQ apigenin 0,0041346 mg/g; LOD quersetin 0.0005547 mg/g dan LOQ quersetin 0.001681 mg/g
