dc.description.abstractBackground: The poguntano plant from the Linderniaceae family has pharmacological activity as a diuretic, anticancer, antipyretic, hepatoprotective, cardioprotective, anti-inflammatory, analgesic and anthelmintic. The contents of poguntano herbs include flavonoids, glycosides, saponins, tannins and steroids. Apigenin and quercetin are flavonoid derivatives in the flavone and flavonol subclasses. The UHPLC method was used to examine apigenin levels in poguntano herb extract. Objective: The purpose of this research was to determine the levels of apigenin and quercetin from the hydroalcoholic extract of the herb poguntano (Picria felterrae Lour.) using the UHPLC method. Method: The research was carried out by making simplicia, characterization of simplicia, making hydroalcoholic extract of Poguntano herb using maceration method, qualitative testing and determination of apigenin and quercetin levels using UHPLC, using the mobile phase aquabides:acetonitrile:methanol (50:30:20) with a flow rate of 0.5 mL/minute, temperature 35ºC, wavelength 350 nm, injection volume 10μL, and C18 (octadecyl) column. The method validation test parameters carried out are linearity, accuracy, precision, limit of detection (LOD), and limit of quantitation (LOQ). Results: The results of the characterization of simplicia water content, water soluble essence content, ethanol soluble essence content, total ash content, insoluble ash content of simplicia acid were respectively 7.98%; 23.55%; 19.31%; 4.19%; 0.71% and water content, total ash content, extract acid insoluble ash content respectively 9.28%; 2.32%; 0.43%. The results of identification using UHPLC of Poguntano herb extract showed that there were apigenin and quercetin compounds at a retention time of 5.562 minutes and 4.017 minutes with levels of apigenin 0.0493 mg/g extract and quercetin 0.5060 mg/g extract. Validation test results obtained % recovery of apigenin 97.81% quercetin 102.95%; RSD apigenin 0.5537% quercetin 1.0668%; LOD apigenin 0.0012 mg/g extract, quercetin 0.0004 mg/g extract; and LOQ apigenin 0.0038 mg/g extract, quercetin 0.0011 mg/g extract. Conclusion: The results of this study showed that the levels of apigenin and quercetin compounds in the hydroalcoholic extract of poguntano herb obtained were 0.0493 mg/g ekstract dan 0.5060 mg/g extract. The UHPLC method used to examine apigenin and quercetin levels has met the validation requirements.en_US
