dc.description.abstract | Background: Mefenamic acid is an analgesic, antipyretic, and anti-inflammatory drug classified as a drug with low solubility and thus causes the speed of dissolution and bioavailability of the drug to be low. Consequently, it is necessary to make preparation of the mefenamic acid nanosuspension to increase the dissolution rate of the drug in order to produce improved analgesic and antipyretic effects, and to reduce the stomach irritation effect due to the use of mefenamic acid.
Objective: To find out the effects of analgesic, antipyretic and irritation on rat administered nanosuspension of mefenamic acid.
Method: A nanosuspension of mefenamic acid made using a nanoprecipitation method by dissolving mefenamic acid in a 0,1 N NaOH solution so as to form a sodium mefenamate solution, and then added a 0,1 N HCl solution while stirring with magnetic stirrer so as to form a mefenamic acid nanoparticle and stabilized with a polymer i.e. gom xanthan thus forms nanosuspension. A mefenamic acid suspension manufacture with a gom xanthan polymer is then carried out. As well as conducted an evaluation of the preparation specifications covering organoleptics. as well as carried out stability evaluation of the preparation covering organoleptic during storage, pH, particle size and dissolution. Further performed an in vivo test using a test minimal i.e., a rat. Observed pain response time (analgetic effect test), temperature observation time (an antipyretic effect test) and stomach of rats macroscopically (the amount of irritation, index of irritation and gastric mucosal damage score) and microscopy (the gastric histopathological result).
Results: The lowest particle size and the fastest dissolution rate are indicated by the preparation of the mefenamic acid nanosuspension formula 21 (N21). Based on the results of a 180 minutes analgesic effect test, the results were successively obtained by nanosuspension N21 (8,37 seconds), nanosuspension N16 (8,12 seconds), suspension S2 (5,9 seconds), and suspension S1 (5,1 seconds). The results of antipyretic effect are obtained results of temperature drop in the 20th minute with nanosuspension N21 and N16 at the earliest i.e. 37,7°C and 37,8°C, and irritation test result was obtained with an average the amount of irritation, index of irritation and gastric mucosal damage score of the nanosuspension groups N21 and N16 respectively, namely nanosuspension N21 (1,33; 0,03%; 1) and nanosuspension N16 (1,67; 0,06%; 1).
Conclusion: Nanosuspension of mefenamic acid gives better analgesic and antipyretic effects, as well as reduce irritation effect on the stomach. | en_US |
