Overview of The Purity and Concentration of DNA Resulting from The Isolation Process Using Branded Reagensia with Homemade Reagensia
Abstract
Background: The DNA isolation process is an important step in various molecular biology and biotechnology research. The purity of the DNA produced is very influential in determining the success of subsequent analyzes such as genetic analysis. Objective: To describe and compare the level of DNA purity resulting from the isolation process using branded reagents with homemade reagents. The isolation process was carried out on samples in the form of human blood by measuring DNA purity through the A260/A280 ratio using a UV-Vis spectrophotometer with ideal values in the range of 1.8 – 2.0. Method: measurement data were analyzed descriptively to determine differences in the level of purity of the DNA produced. Results: research results show that branded reagents produce DNA with a more consistent and ideal level of purity. However, homemade reagents show competitive results and are almost equivalent in purity to branded reagents. Conclusion: this study concludes that homemade reagents can be a more economical alternative and can be developed for future research needs. It is hoped that this research can become a reference in developing more efficient DNA isolation methods to achieve optimal DNA quality.
