    Abstract
    Background. The prevalence of free radical-related diseases is continuously rising, driving the advancement of research and the usage of herbal medicines, including Moringa oleifera, as antioxidants. However, research on combining herbal and conventional antioxidants to reduce malondialdehyde (MDA) levels is still limited. Objective. To compare the effects of combined herbal and conventional antioxidants with single formulations. Methods. This research analyzed the characterization of simplicia, phytochemical screening of moringa leaves water extract, and serum MDA concentration using the ELISA assay. A total of 42 rats were divided into seven groups: a negative control, positive control, and five treatment groups. The positive control was exposed to lead only for 7 days, treatment groups were exposed to lead for 7 days followed by 14 days of antioxidant supplementation using Moringa leaves water extract (200 mg/kgBW), vitamin C (7.2 mg/200 gBW), collagen (5.4 mg/200 gBW), and their combinations. Statistical analyses were performed using SPSS. Data were then analyzed using SPSS, utilizing the Kruskal-Wallis test and the Mann-Whitney test. A p-value of < 0.05 was considered statistically significant. Results and Discussion. The water content of Moringa leaves simplicia was 8.64%, with water-soluble extract at 9.35% and ethanol-soluble extract at 14.17%. There is a significant difference in MDA levels among groups (p < 0.001). The MDA levels in the Moringa leaf extract (MLE) group compared to the combination of MLE and collagen (21.32 ± 0.95 nmol/mL vs. 14.65 ± 0.66) showed a significant difference (p = 0.004) Conclusion. The moringa leaf extract has antioxidant activity, and the combination of moringa leaf extract with collagen and vitamin C is not better than collagen and vitamin C monotherapy in reducing MDA levels in an oxidative stress animal model.
