dc.description.abstractThe objective of this research was to determine the growth response of black cumin shoots (Nigella sativa L.) to the application of a combination of the plant growth regulators NAA and BAP under in vitro conditions. This research was conducted in the Biotechnology Laboratory of the Faculty of Agriculture, Universitas Sumatera Utara, from April 2024 to August 2024. The study employed a Completely Randomized Design (CRD) with two treatment factors, each with three replicates. The first treatment factor was the concentration of NAA (0 ppm, 0.5 ppm, and 1 ppm), while the second was the concentration of BAP (0 ppm, 1.5 ppm, and 3 ppm). Observed parameters included the percentage of explants forming shoots (%), the percentage of explants forming callus (%), callus texture, callus color, shoot emergence time (days), shoot length (cm), and the number of shoots. The results showed that NAA concentration had no significant effect on shoot emergence time, shoot length, or the number of shoots but significantly affected the percentage of explants forming callus. BAP concentration significantly influenced shoot emergence time, shoot length, and the number of shoots. The interaction between NAA and BAP concentrations did not significantly affect shoot emergence time, shoot length, or the number of shoots but significantly affected the percentage of explants forming callus.en_US
