Identifikasi DNA Barcoding dan Status Kekerabatan Rajungan (Portunus sp.) di Perairan Pantai Labu

Abstract

Blue Swimming Crab (Portunus sp.) is one of the high economic value fishery commodities found in Pantai Labu Waters. However, information on crab species caught in this area is still limited, so accurate identification is needed to support conservation and fisheries resource management efforts. This study aims to identify crab species (Portunus sp.) caught in Pantai Labu Waters using the DNA barcoding method and analyze their kinship status. Crab samples were collected from two locations using random sampling method and analyzed molecularly with Polymerase Chain Reaction (PCR) technique and DNA sequencing. The results showed that three species of crab were identified, namely Portunus pelagicus, Monomia gladiator, and Charybdis variegata. Phylogenetic analysis using the Neighbor-Joining tree method with the Kimura 2-Parameter model shows the relationship between species based on differences in genetic distance. The results of this study provide basic information that can be used in efforts to manage and conserve crabs in the waters of Pantai Labu and as a reference in the development of sustainable crab aquaculture.
