Research was conducted with the aim of obtaining lactic acid bacteria (LAB) species from dengke naniura that have the ability to reduce cholesterol. The research was carried out in several stages, each stage is done with the method standardised procedures with the reference. LAB isolates isolated were indicated by the presence of a clear zone around the colony. Identification results of 15 isolates showed the characteristics of LAB, namely round or rod-shaped cells, negative catalase, non-motile, and Gram positive. The isolates tested were coded RS-1 to RS-15. All isolates able to grow on media containing 0.4% oxgall and could live on media with pH of 3.0. All LAB isolates showed the ability to reduce cholesterol by 34%–65%. Four isolates with the highest cholesterol-reducing ability were selected for molecular identification: two round (RS-3 and RS-9) and two rod (RS-12 and RS-14). The results of identification using the 16S rRNA gene show that isolates RS-3 and RS-9 have in common with Pediococcus acidilactici, RS-12 with Lactobacillus plantarum, and isolate RS-14 with Lactobacillus paracasei. Whole genome sequence identification was carried out on RS-9 and RS-12, and it was found that isolate RS-9 has similarities with Pediococcus pentosaceu ATCC 33316 and RS-12 with Lactiplantibacillus plantarum subsp. plantarum DSM 20174. In silico genomic prediction test using RAST, it was found that Pediococcus pentosaceus RS-9 and Lactiplantibacillus plantarum subsp. plantarum RS-12 have enzyme-producing genes that play a role in lowering cholesterol called Bile Salt Hydrolase enzyme. Conclusions is dengke naniura contains LAB that able to lower cholesterol.
