    The Effect of Nigella Sativa Extract on the Growth of Pseudomonas Aeruginosa in Secretions of Patients with Chronic Suppurative Otitis Media (CSOM)

    Abstract
    Introduction : CSOM is a chronic inflammation of the middle ear associated with perforation of the tympanic membrane and otorrhea, which often leads to hearing loss. The widespread and often improper use of antibiotics has contributed to antibiotic resistance. Nigella sativa has been traditionally used for its medicinal properties and has shown potential as an antibacterial agent. Objective: This study aims to evaluate the antibacterial effect of Nigella sativa extract on the growth of Pseudomonas aeruginosa in the secretions of patients with CSOM. Methods: The study used an in vitro experimental design with the disc diffusion and well diffusion methods. Nigella sativa seeds were extracted into eight different concentrations: 50%, 75%, 100%, 0.02 mg/mL, 2 mg/mL, 3 mg/mL, 4 mg/mL, and 5 mg/mL, and compared with a positive control (ofloxacin 5 mcg) and a negative control (1% DMSO). Results: Statistical analysis using the Kruskal-Wallis test followed by Dunn’s post-hoc test showed a significant difference in the inhibition zone diameter using the disc diffusion method between the negative control group and the positive control group (p<0.001), and between the positive control group and all Nigella sativa extract groups (p<0.001). Using the well diffusion method, a significant difference in inhibition zone diameter was found between the negative control and the Nigella sativa 4 mg/mL and 5 mg/mL extract groups (p<0.001). No significant difference was found between the negative control and the Nigella sativa extract at concentrations of 0.02 mg/mL, 2 mg/mL, and 3 mg/mL, indicating a minimum inhibitory concentration (MIC) of 4 mg/mL and 5 mg/ml. Conclusion: Nigella sativa extract shows antibacterial activity against Pseudomonas aeruginosa. Keywords: CSOM;Nigella sativa;Pseudomonas aeruginosa;antibiotic resistance
