The main cause of dental caries is the pathogenic microorganism Streptococcus mutans. The most practical way to prevent dental caries is to inhibit and kill S.mutans bacteria. This study aims to determine whether tomato fruit extract (Solanum lycopersicum L.) can inhibit the growth of S.mutans. The extract was obtained by maceration method with concentrations of 3.125%, 6.25%, 12.5%, 25%, 50%, Chlorhexidine gluconate 0.2% as positive control and Dimethyl as negative control. The purpose of this study was to determine the effectiveness of tomato fruit extract against the growth of Streptococcus mutans bacteria with the type of laboratory experimental research and post-test only control group design in vitro. Antibacterial testing using disc paper diffusion method. The analysis used normality test and homogeneity test, the results of which showed that the data were not normally distributed and not homogeneous. Followed by Kruskall-Wallis and Mann-Whitney tests which showed that there was a zone of inhibition and there were significant differences between all treatment groups (p<0.05). It can be stated that tomato fruit extract (Solanum lycopersicum L.) effectively inhibits the growth of Streptococcus mutans bacteria with inhibition zones at a concentration of 3.125%: 7.58 mm; 6.25%: 10.47 mm; 12.5%: 12.79 mm, 25%: 15.62 mm; 50%: 18.04 mm.
