dc.description.abstractIntroduction : Regenerative endodontic treatment has been suggested as an alternative biologically based treatment for necrotic adult permanent teeth with closed and well-formed apex. The use of appropriate irrigation solutions can release TGF-β1 stored in dentin, thereby contributing to the success of regenerative endodontic treatment. Therefore, irrigation solution of Quantumdots (QDs) extract lerak 4%, extract lerak 12.5% and 17% EDTA is necessary to stimulate the release of TGF-β1 stored in dentin which contributes to the success of regenerative endodontic treatment. The QDs extract lerak 4% were subjected to characteristic tests, namely density, viscosity, flow rate, UV light, UV-Vis, Photoluminescence (PL), and HR-Tem which proved that QDs extract lerak 4% were successfully synthesized. In addition, the pH of QDs extract lerak 4% and extract lerak 12.5% was measured, which showed that both solutions had an acidic pH, so they could stimulate the release of TGF-β1. Materials and methods : Twenty-seven single root premolars underwent root canal treatment. Root canals were prepared with V-Taper Gold rotary file system to size #25.08. Teeth that had been prepared were irrigated and divided into 3 groups, namely 1.5% NaOCl + 4% lerak extract QDs, 1.5% NaOCl + 12.5% lerak extract, and 1.5% NaOCl + 17% EDTA then agitate with ultrasonic for 1 minute. The irrigation solution was collected in a container and tested for TGF-β1 using the Elisa Kit. The data obtained were subjected to the Kruskal Wallis test and continued with the Mann-Whitney test. Results : The results of the Kruskal Wallis test showed significant differences (p<0.05) between the three groups. However, the Mann-Whitney test showed that the use of 1.5% NaOCl irrigation solution + 4% lerak extract QDs with 1.5% NaOCl + 17% EDTA did not show a significant difference. Conclusion : From this research it can be concluded that irrigation using 1.5% NaOCl + 4% lerak extract QDs has the same effectiveness as 1.5% NaOCl + 17% EDTA irrigation solution on the release of TGF-β1.en_US
