Background: Ibuprofen is a non-steroidal anti-inflammatory drug (NSAID). One
of the disadvantages of NSAIDs can cause stomach ulcers. Thus, ibuprofen Solid
SEDDS preparation was formulated to reduce the gastric irritating effect of NSAID
class drugs and a comparison was made with Liquid SEDDS.
Objective: To formulate, characterize, test the gastric irritation reduction and
antiinflammatory effects of ibuprofen Solid SEDDS preparation in rats.
Methods: Formulation of ibuprofen Solid SEDDS preparation by adsorbtion to
solid carrier method with adsorbent variation (aerosil, talcum, lactose). Then the
best Solid SEDDS formula was taken to compare characterization with Liquid
SEDDS including organoleptic, pH, particle size, zeta potential, content and
dissolution. Furthermore, the stability test of the preparation includes organoleptic,
drug content, particle size, pH and dissolution. Continued testing of anti
inflammatory effects, and gastric irritation effects using rats consisting of 4 groups.
Results: From the results of the study, aerosil was found to be the best adsorbent
for the Solid SEDDS formula. From the characterization results obtained particle
size of 461.91 ± 19.55, content of 104.82 ± 5.28%, and zeta potential of -38.1 ±
0.17 mV. From the dissolution results of Solid SEDDS at the 60th minute, it reached
88.25% in phosphate buffer pH 7.2. In testing the anti-inflammatory effect, Solid
SEDDS produced the same effect as ibuprofen Liquid SEDDS, while for testing the
gastric irritation effect, ibuprofen Solid SEDDS proved to reduce the gastric
irritation effect significantly compared to ibuprofen suspension but produced the
same effect as Liquid SEDDS.
Conclusion: Based on the results of the research, it can be concluded that ibuprofen
can be formulated in Solid SEDDS dosage form by adsorbtion to solid carrier
method, Solid SEDDS ibuprofen has the same anti-inflammatory effect and gastric
irritation reduction effect as Liquid SEDDS but significantly different from
ibuprofen suspension.
