Background : Free radicals are reactive compounds that can cause oxidative stress and contribute to various degenerative diseases such as cancer, diabetes and cardiovascular disease. Antioxidants play an important role in neutralizing free radicals and preventing cell damage. One potential source of natural antioxidants is gagatan harimau leaves (Ampelocissus thyrsiflora (Blume) Planch) which is traditionally used to improve stamina and health. However, until now there are still few studies that examine its antioxidant potential scientifically, especially with different testing methods. Therefore, this study aims to evaluate the antioxidant potential of ethanol extract of gagatan harimau leaves using DPPH, ABTS, and FRAP methods.
Objective : This study aims to determine the IC50 value of antioxidant potential of ethanol extract of gagatan harimau leaves using DPPH, ABTS, and FRAP methods and compare their antioxidant potential.
Methods : This study used maceration extraction method with 96% ethanol solvent. Antioxidant potential was tested using DPPH, ABTS, and FRAP methods with UV-Vis spectrophotometry. Data were analyzed statistically with One way anova test at 95% confidence level to determine the significance of differences between methods.
Result : The results showed that the IC50 value of ethanol extract of tiger crow leaf with DPPH method was 19.9372 ± 0.02 µg/mL, FRAP method was 32.2448 ± 0.11 µg/mL, and ABTS method was 80.6468 ± 0.21 µg/mL. DPPH method showed very strong antioxidant activity, FRAP method also showed very strong activity but lower than DPPH, while ABTS method only showed strong activity. Statistical tests showed significant differences (p < 0.05) between methods.
Conclusion : Ethanol extract of gagatan harimau leaves has high antioxidant potential. The highest antioxidant potential was obtained by DPPH method, followed by FRAP method, and finally ABTS method. These results show that the testing method can affect the results of antioxidant activity of an extract.
