    Abstract
    Background: Beet tubers (Beta vulgaris Lindl.) are known to contain bioactive compounds such as flavonoids that have potential as antioxidants, anti-inflammatory and antibacterial. Among these compounds, quercetin, naringenin, and kaempferol are of interest because they have various health benefits. Objective: This study aimed to analyze the content of quercetin, naringenin, and kaempferol in beet tubers using Ultra performance liquid chromatography (UPLC) method. Methods: Beetroot samples were extracted using maceration method with methanol solvent pro analysis. The analysis was performed using UPLC system with MRM detector (Acquity UPLC® BEH C18 1.7 µm (2.1 x 100 mm) column, mobile phase A is a mixture of 0.1% formic acid in water and B is a mixture of 0.1% formic acid in acetonitrile (using gradient elution), the test was performed using Multiple reaction monitoring (MRM) detector at the optimal wavelength, and the retention time for each component was determined based on the standard. Results: The results of this study showed that the three compounds, namely quercetin, naringenin, and kaempferol, were successfully identified using the MRM detector and the following specific parameters. Quercetin was detected with a mobile phase ratio of A(69.7):B(30.3), retention time of 4.79 min, and analysis wavelength at 360 nm, with a measured level of (0.002021±0.000357) mg/g. Naringenin was identified with a mobile phase ratio of A(50):B(50), a retention time of 5.85 minutes, and an analysis wavelength at 280 nm, with a level of (0.028274±0.000190) mg/g. Meanwhile, kaempferol was detected with a mobile phase ratio of A(50):B(50), a retention time of 6.14 min, and an analysis wavelength at 280 nm, with a concentration of (0.003637±0.000215) mg/g. Conclusion: UPLC MS/MS analysis showed that beetroot contains quercetin, naringenin, and kaempferol compounds in certain amounts. The three compounds were detected with good peak resolution and clearly separated retention times.
