    Formulation of Hydrogel from Ethanol Extract of Catharanthus roseus L. Leaves and Test of Wound Healing Effectiveness in Rats

    Abstract
    Background: Cathranthus roseus leaves contain alkaloid, tannin and flavonoid compounds that can play a role in accelerating wound healing. Hydrogel preparations can accelerate wound healing by rehydrating tissues without causing tissue trauma to the wound bed. Objective: Formulate hydrogel preparations of Catharanthus roseus leaves ethanol extract and evaluate the effectiveness of incision wound healing in rats. Method: The study was conducted experimentally using 25 white rats divided into 5 groups, namely negative control (hydrogel base without extract), positive control (Hansaplast wound ointment), and the other 3 groups were given hydrogel preparations of Catharanthus roseus leaves ethanol extract at concentrations of 0.5; 1; and 2%. The preparation was evaluated and tested for its wound healing effectiveness on the backs of rats that were made a 2 cm incision wound using a scapel. The wound was cleaned using sterile NaCl solution and the preparations were given according to the group. Percent wound reduction was observed for 10 days. Results: The hydrogel preparation of Catharanthus roseus leaves ethanol extract is homogeneous, brownish yellow in color, smells typical of the extract, semisolid, pH 5,15-5,66, viscosity 5659,03-61114,3 cP, spreadability 5,03-6,47 cm, stable in room temperature storage for 12 weeks. The hydrogel with ethanol extract of Catharanthus roseus leaves at concentrations of 0.5%; 1%; and 2% resulted in wound reduction percentages of 77.85±1.1%; 86.46±3.24%; and 100±0% on day 11, respectively. All three concentrations had significant differences with the negative control (p < 0,05). Conclusion: The ethanol extract of Catharanthus roseus leaves can be formulated in a hydrogel preparation, stable in room temperature storage for 12 weeks and meets the physical evaluation requirements. The hydrogel preparation of ethanol extract of Catharanthus roseus leaves is effective in healing wound.
