This study aims to determine the effect of Casein hydrolysate concentration and
BAP on the growth of red barangan banana in vitro. This research was conducted
at the Laboratory of Plant Biotechnology, Faculty of Agriculture, University of
North Sumatra in November 2024 - February 2025. This study used a complete
randomized design (CRD) method with two factorials. The first factor is the
concentration of Casein hydrolysate (C): C0 (0 mg/l), C1 (300 mg/l), and C2 (600
mg/l). The second factor is the concentration of Benzyl Amino Purine (BAP): B0:
(0 mg/l), B1: (2.5 mg/l), and B2: (5 mg/l). The parameters observed in this study
include, percentage of living explants (%), age of leaflets, number of shoots, length
of shoots, number of roots, and root length. The results showed that the combination
of Casein hydrolysate and BAP significantly affected the root length parameter.
The best treatment for root length was obtained from the combination of Casein
hydrolysate 600 mg/l with BAP 2.5 mg/l.
