Background: Jeruju (Acanthus ilicifolius Linn.) contains efficacious compounds such as alkaloids, flavonoids, steroids, and tannins that have biological activities included antibacterial, anti-inflammatory, and antioxidant that can accelerate wound healing. The use of chitosan is able to inhibit the growth of pathogenic bacteria so that the combination of these two ingredients is able to increase antibacterial activity which plays an important role in the healing of burn wound. Objective: This study aimed to test the wound healing effect of hydrogels mixed of EEDJ and chitosan on male rabbits induced burn wounds. Method: This test used male rabbits which were divided into 5 treatment groups, namely hydrogels mixed of EEDJ and chitosan with varying extract concentrations (2.5%; 5%; 10%), hydrogel of EEDJ 10%, and chitosan hydrogel, as well as 2 control groups, namely hydrogel base (vehicular control) and Bioplacenton® Gel (positive control). The test animals were made burn wounds using iron plate with a diameter of 24 mm that had been heated on hot plate at a temperature of 150°C for 5 minutes. The test animals were given treatment and observed burn wounds for 21 days and then the area of burn wounds was measured using ImageJ application. Results: The results of this test showed that the hydrogels mixed of EEDJ and chitosan had a wound healing effect, marked by a significant differences (p < 0.1) with hydrogel base (vehicular control). The hydrogel with extract concentration of 2.5% is the most effective hydrogel in healing burn wounds with a total AUC value of the burn wound area shrinkage percentage of 562.29±256.83%. Conclusion: The hydrogels mixed of EEDJ and chitosan had a wound healing effect on male rabbits induced burn wounds.en_US
