    Abstract
    Background: Melasma is a condition of acquired hypermelanosis, in the form of symmetrical macules, light brown to dark brown generally in areas exposed to ultraviolet (UV) light with a predilection on the cheeks, forehead, upper lip area, nose and chin. The main goal of melasma therapy is to completely disguise or eliminate hyperpigmentation, reduce the size of the lesion, and prevent recurrence. An in vitro study on avocado seed extract showed its effect as a potent tyrosinase inhibitor, so that natural depigmenting agents from herbal ingredients such as avocado seed extract may provide opportunities as an effective method in treating melasma. Objective: To determine the effect of 1% avocado seed extract nanoemulgel (Persea americana Mill.) on melasma Methods: Pre-experimental clinical trial research with a pretest-posttest design on 30 melasma subjects. Diagnosis is made through history and clinical examination. The assessment was conducted using the Melasma Area Severity Index (MASI), mexameter and skin analyzer which were assessed before and after administration of 1% avocado seed extract nanoemulgel (Persea americana Mill.). Evaluation was conducted every 2 weeks, side effects were observed and noted during the study. Friedman test was carried out to see the differences before and after treatment with a p value <0.05 considered significant. Result: The characteristics of the study subjects showed that the majority were aged 40-50 years (46.67%) with an average age of 45.03±4.63. The application of 1% avocado seed extract nanoemulgel showed a significant difference in MASI scores, melanin index, RGB spot, and UV pigmentation (p<0.05). In this study, no side effects were found after the application of 1% avocado seed extract nanoemulgel. Conclusion: The application of 1% nanoemulgel of avocado seed extract (Persea americana Mill.) can improve the appearance of melasma Keywords: melasma, melasma area severity index, melanin index, avocado seed, Persea americana Mill.
