Effect of Concentration and Duration of Colchicine Immersion on The Growth of Common Bean Embryos (Phaseolus vulgaris L.) In Vitro
Abstract
This study aims to determine the effect of concentration and duration of colchicine immersion on the growth of common bean embryos. This research was conducted at the Alifa Agricultural Research Center (ALIFA-ARC) Laboratory and the Plant Biotechnology Laboratory of the Faculty of Agriculture, University of North Sumatra, from September to November 2024. This study used a Completely Randomized Design method with two treatment factors. The first factor is the concentration of colchicine consisting of 0 ppm (control), 50 ppm, 100 ppm and 150 ppm. The second factor is the duration of colchicine immersion consisting of 2 hours, 4 hours and 6 hours. The parameters observed were the percentage of living explants, plantlet length, number of leaves, number of buds, wet weight, stem diameter, stomata size and number of stomata. The results showed that the concentration of colchicine did not have a significant effect on all observation parameters. Colchicine soaking time of 2 hours had a significant effect on planlet length, number of leaves, number of buds, wet weight and stem diameter. The interaction between the two treatments had no significant effect on all observation parameters.
