dc.description.abstractSiporang rice (Oryza sativa L.) from South Tapanuli Regency, North Sumatra, exhibits resistance to blast disease. This resistance is presumed to be associated with the presence of R genes involved in the plant defense system, one of which is the pi-ta gene. This study aimed to isolate and sequence a fragment of the pi-ta gene from Siporang rice to provide genetic information for plant breeders. The research stages included genomic DNA isolation from Siporang rice, DNA amplification using pi-ta-specific primers, and sequence analysis using NCBI, BLASTn, and MEGA12. Based on the results, the pi-ta gene fragment isolated had a size of 774 bp. BLASTn analysis showed that the gene fragment successfully isolated from Siporang rice (Oryza sativa var. Siporang L.) was confirmed to be the pi-ta gene, with a 99.85% identity to the pi-ta gene sequence from rice cultivar Banks (GQ918477.1). Phylogenetic analysis using the Maximum Likelihood method produced a dendrogram with an evolutionary distance value of 0.16 compared to five other pi-ta gene sequences retrieved from GenBank. The sequence information obtained is expected to be useful for further studies in the development of blast-resistant rice cultivars with optimal yield potentialen_US
