dc.description.abstract | This study aims to determine the effect of colchicine soaking concentration and duration on the growth of long bean embryos (Vigna sinensis L.) in vitro. This research was conducted at the Alifa Agricultural Research Center (ALIFA-ARC) Laboratory from October 2024 to January 2025. This study used a Factorial Completely Randomized Design (RAL) method with two factors. The first factor was colchicine concentration consisting of 0 ppm, 50 ppm and 100 ppm. The second factor is colchicine soaking time consisting of 0 hours, 4 hours and 6 hours. The parameters observed were the percentage of live explants, planlet length, number of leaves, number of buds, wet weight, stem diameter, stomatal size and number of stomata. The results showed that the administration of colchicine had a significant effect on the parameters of the number of leaves, number of buds, wet weight and stem diameter in the treatment of 50 ppm concentration (K1). In the provision of colchicine soaking time has a real effect on the parameters of planlet length, number of leaves, number of buds, wet weight and stem diameter in the treatment of 4 hours immersion (L1). Based on the results showed that the administration of colchicine with a concentration of 50 ppm with a soaking time of 4 hours can increase the growth of broad bean embryos in vitro. | en_US |
