dc.description.abstractBackground : Gotu kola (Centella asiatica (L.) Urban) is a plant that has benefits in wound healing with its main content is triterpenoids which include asiaticoside, centeloside, madecoside and asiatic acid. It was known that polyphenolic compounds had the potential to enhance platelet aggregation and accelerate coagulation. Therefore, a comparison was conducted between the Ethanol Extract of Gotu Kola Herb (EEHP) and the Polyphenol-Rich Extract of Gotu Kola Herb (EKPHP). Objective : This study aims to determine the difference between Ethanol Extract of Gotu kola Herb (EEHP) and Polyphenol-Rich Extract of Gotu Kola Herb (EKPHP) on hemostasis activity. Methods : The extraction process used the maceration method for the ethanol extract of gotu kola herb and the polyphenol-rich extract of gotu kola herb, followed by the fractionation method to obtain the polyphenol-rich extract. Subsequently, hemostasis activity testing was conducted in vitro by Lee-White method to determine coagulation time with stopwatch, the Eustrek method to observe hemostasis activity microscopically, and the platelet aggregation analysis method by a UV-Vis spectrophotometer. Results : The normal blood coagulation time was 2.55 ± 0.12. The EDTA + EEHP (0.5%; 1%; 2%) group had a coagulation time of 120 ± 0,00 (not clotting). The coagulation times for EEHP 0.5%, EEHP 1%, and EEHP 2% were 1.57 ± 0.06, 1.63 ± 0.04, and 1.74 ± 0.07, respectively. Meanwhile, the EKPHP test group showed better results, with coagulation times of 1.23 ± 0.14 for EKPHP 0.5%, 1.63 ± 0.04 for EKPHP 1%, and 1.74 ± 0.07 for EKPHP 2%. Microscopically, both EEHP and EKPHP stimulated platelet aggregation and coagulation formation. The percentage of transmission measured by a UV-Vis spectrophotometer showed that EEHP at 50% to 80% had transmission values ranging from 46.6% to 63.7%, whereas EKPHP demonstrated better results, with transmission values ranging from 56.7% to 72% at 50% to 80%. Conclusion : The polyphenol-rich extract of gotu kola herb effectively accelerated coagulation time, enhanced hemostasis activity, and increased platelet aggregation compared to the ethanol extract of gotu kola herb.en_US
