Isolation And Identification Of Flavonoid Compounds From The Bark Of Api – Api Mangrove (Avicennia Marina (Forssk.) Vierh.)

Isolation and identification of flavonoid compounds from the bark of the api-
api mangrove (Avicennia marina (Forssk.) Vierh.) have been carried out through
several stages, namely maceration extraction with methanol solvent, fractionation
with ethyl acetate solvent, fractionation with n-hexane solvent to obtain methanol
extract. Then evaporated until all methanol has evaporated. The concentrated
methanol extract was analyzed by TLC, then separated by column chromatography
with silica gel as the stationary phase and chloroform: ethyl acetate (90:10; 80:20;
70:30; 60:40; 50:50; 40:60; 30:70; 20:80; 10:90) v/v as the mobile phase. The fourth
fraction was purified by Preparative TLC with silica gel as the stationary phase and
chloroform: ethyl acetate (20:80) v/v as the mobile phase. The pure compound
obtained was in the form of a yellow amorphous solid of 5.2 mg with an Rf value of
0.55. Furthermore, the compounds obtained were analyzed using UV-Visible
Spectrophotometers with the results obtained 2 bands namely band I with maks 371 nm
with an absorbance of 0.33871 and band II with maks 271 nm with an absorbance of
0.31856, Infrared Spectrophotometer (FT-IR) analysis with the results obtained the
presence of functional groups O-H Stretching, C-H Stretching, C=O ketone Stretching,
C=C aromatic Stretching, C-H sp3 Bending, C-O Stretching, C-H sp2 Bending out-of-
plane, with wave numbers respectively 3425.58, 2956.87, 1726.29, 1598.99, 1381.03,
1286.52, 742.59 cm-1
and Proton Nuclear Magnetic Resonance Spectrophotometer
analysis (1H-NMR) with the results obtained the presence of protons H-6, H-8, H-2’,
H-5’, H-6’, OCH3 and no protons at C-3. Spectroscopic data shows that the isolated
compound is a flavonoid compound of the flavonol group.
