Isolation and Identification of Flavonoids Compounds from The Bark or Jirak (Eurya acuminata DC.)
Abstract
Isolation and identification of flavonoid compounds from the bark of Jirak
(Eurya acuminata DC.) has been done through several stages, namely maceration
extraction with methanol solvent, fractionation with ethyl acetate solvent,
fractionation with n-hexane solvent until the methanol layer is obtained and then
evaporated until all the methanol evaporates. The methanol concentrated extract was
then analyzed by KLT, then separated by column chromatography with silica gel as
stationary phase and chloroform: ethyl acetate (90:10; 80:20; 70:30; 60:40; 50:50;
40:60; 30:70; 20:80; 10:90) v/v as mobile phase. The compound obtained was a
yellow amorphous solid weighing 5 mg with an Rf = 0.44 with eluent chloroform:
ethyl acetate (20:80) v/v. Based on the analysis with UV-Vis spectroscopy with
methanol solvent showed two absorption maximum wavelength (λmax) 318 and 279
nm. Analysis with FT-IR spectroscopy showed the presence of functional groups O-H
stretching, C-H sp3 stretching, C=O ketone with stretching vibration, C=C
aromatic, C-H bending, C-O, C-H sp2 bending out-of-plane, with wave numbers
respectively 3425.57 cm-1, 2922.15 cm-1, 1729.21 cm-1, 1589.34 cm-1, 1382.96 cm
1, 1259.52 cm-1, 790.81 cm-1. The Proton Core Magnetic Resonance (1H-NMR)
spectrum showed the presence of protons H-6 & H-8, H-3'& H-5', H-2' & H-6', and
OCH3. From the spectrum analysis, it can be concluded that the isolated compound
is a flavonoid compound of flavone group.
