dc.description.abstractIntroduction : Vital pulp therapy (VPT) requires biocompatible materials that support pulp tissue regeneration through angiogenesis on dentinogenesis activity. One important indicator of this regeneration process is the viability of human Dental Pulp Stem Cells (hDPSCs) and the concentration of Vascular Endothelial Growth Factor (VEGF). Nano chitosan-silica-calcium carbonate biocomposite (Biokomposit nKSC) was developed as an alternative pulp capping material such as Biodentin. The purpose of this study was to analyze the effect of Biokomposit nKSC on hDPSCs cell viability and VEGF concentration compared to Biodentin. Material and Method : In vitro studies were conducted on hDPSCs cells exposed to nKSC and Biodentin. Cell viability tests were performed using MTT assay, and VEGF concentration measurements using ELISA at 24 and 72 hours. Data were statistically analyzed using Wallis and Pos Hoc Mann-Whitney for cells viability; ANOVA and Post Hoc LSD tests for VEGF concentration. Results: The highest hDPSCs cell viability at 72 hours was shown by the Biokomposit nKSC group (0.981 ± 0.013), higher than Biodentin (0.964 ± 0.011) and control (0.957 ± 0.017). VEGF concentration was also significantly higher in the Biokomposit nKSC group (60.31 ± 2.17 pg/mL) compared to Biodentin (56.21 ± 2.03 pg/mL) and control (52.68 ± 1.91 pg/mL) at 72 hours (p<0.05). Conclusion: Biokomposit nKSC showed higher cell viability and VEGF concentration than Biodentin, thus having high potential as an alternative biomaterial in vital pulp Therapy.en_US
