    Abstract
    Background: Breast cancer is the leading cause of death among women in Indonesia. Conventional therapies often cause serious side effects and incur high costs. Saurauia vulcani Korth. (Pirdot) contains bioactive compounds with potential as anticancer agents. Objective: To evaluate the anticancer activity of ethanolic extract and three fractions of Pirdot leaves (n-hexane, ethyl acetate, and residual fraction) against MCF-7 breast cancer cells. Methods: LC-HRMS was performed on the ethanolic extract of Pirdot leaves to identify compounds with potential anticancer activity. The leaves were extracted using ethanol through maceration, then fractionated with n-hexane and ethyl acetate via successive solvent fractionation. The samples were tested at various concentrations for anticancer activity using the MTT assay. Results: LC-HRMS analysis revealed that the ethanolic extract of Pirdot leaves (EEDP) contains bioactive compounds with potential anticancer effects, including oleanolic acid, ursolic acid, and quercetin. The lowest percentage of cell viability in EEDP was 55.76% at 100 µg/mL; for the n-hexane fraction, 41.3% at 400 µg/mL; for the ethyl acetate fraction, 40.55% at 50 µg/mL; and for the residual fraction, 39.44% at 800 µg/mL. Conclusion: The ethanolic extract, n-hexane fraction, ethyl acetate fraction, and residual fraction of Saurauia vulcani Korth. leaves showed no anticancer activity against MCF-7 breast cancer cells.
