Isolation and Identification of Flavonoid Compounds from Key Lime Leaves (Citrus aurantiifolia (Christm.)Swingle)
Abstract
Isolation and identification of flavonoid compounds from Lime Leaves (Citrus aurantiifolia (Christm.)Swingle) has been carried out through the maceration extraction stage with methanol solvent. The methanol extract dissolved with ethyl acetate was separated and concentrated. The concentrated extract of ethyl acetate was dissolved with methanol and partition extracted with n-hexane. The methanol layer is then evaporated until all the methanol has evaporated. The concentrated methanol extract was then analyzed by TLC, then separated by column chromatography with silica gel as the stationary phase and chloroform: ethyl acetate (90:10; 80:20; 70:30; 60:40; 50:50; 40:60; 30: 70; 20:80; 10:90) v/v as mobile phase. The second fraction was chosen to be purified by preparative TLC with silica gel as the stationary phase and chloroform: ethyl acetate (20:80) v/v as the mobile phase. The pure compound obtained was a yellow amorphous solid of 6 mg with a value of Rf = 0.56. Next, the compounds obtained were explained using a UV-Visible Spectrophotometer with the results obtained in 2 bands, namely the first band with a wavelength of 327 nm and the second band with a wavelength of 287 nm, Infrared Spectrophotometer Analysis (FT-IR) with the results obtained that there are functional groups O-H Stretching, O-H, C=O ketone, C=C aromatis, C-O-C, C-OH,, with successive wave numbers 3427,51 cm-1, 1689,64 cm-1, 1506,85cm-1, 1230,58cm-1, 1074,35cm-1. and Proton Nuclear Magnetic Resonance Spectrophotometer (1H-NMR) analysis with the results obtained of the presence of H-6 & H-8, H-2’dan H-6’.Spectroscopic data shows that the isolated compound is a flavonoid compound isoflavon group.
