    Abstract
    This study aims to determine the effect of the combination of natural PGR (coconut water) and 2,4D (Dichlorophenoxy-acetic acid) in MS media on callus induction of grape plants (Vitis vinifera L.). This study was conducted in the Plant Tissue Culture Laboratory, Faculty of Agriculture, University of North Sumatra with an altitude of ± 25 meters above sea level. In January to May 2025. The experimental design used in this study was a factorial completely randomized design (CRD) consisting of 2 treatment factors with each treatment of 6 replications, namely 0 ppm 2.4D 0% Coconut Water (D0P0), 0.5 ppm 2.4D 0% Coconut Water (D1P0), 1 ppm 2.4D 0% Coconut Water (D2P0), 1.5 ppm 2.4D 0% Coconut Water (D3P0), 0 ppm 2.4D 5% Coconut Water (D0P1), 0.5 ppm 2.4D 5% Coconut Water (D1P1), 1 ppm 2.4D 5% Coconut Water (D2P1), 1.5 ppm 2.4D 5% Coconut Water D3P1, 0 ppm 2.4D 10% Coconut Water (D0P2), 0.5 ppm 2.4D 10% Coconut Water (D1P2), 1 ppm 2.4D 10% Coconut Water (D2P2), 1.5 ppm 2.4D 10% Coconut Water D3P2, the parameters observed in this study include the percentage of explant growth (%), the percentage of explant contamination (%), the percentage of callus formation, the day of callus emergence (%), callus color, callus texture, callus diameter, callus length, callus fresh weight (%). The results of the study showed that the interaction of giving various concentrations of 2.4D and Coconut Water had a significant effect on callus growth but did not have a significant effect on all parameters
