The Effect of the Concentration of Plant Growth Regulators NAA And BAP on the Germination of Sweet Orange Seeds of the Brastepu Variety In Vitro
Abstract
Oranges are one of the plants that are widely cultivated in the world. The
problem of the production of Berastagi sweet oranges includes pests and diseases,
weather factors, less fertile soil quality and non -optimal nutrition and lack of
appropriate care causes the quality and production of oranges decreases. Efforts
that can be made to maintain superior nature of sweet orange plants are related
to the propagation of tissue culture. This study aims to determine the effect of the
concentration of NAA and BAP growth regulators on germination of sweet orange
seeds in vitro. This study was conducted at the Plant Biotechnology Laboratory,
Faculty of Agriculture, University of North Sumatra from October 2024 to March
2025. The method used was a complete random design (RAL) with two treatment
factors. The first factor is the NAA concentration with a level of 0, 1.5 ppm, 2
ppm, and 2.5 ppm and the second factor is the concentration of BAP with a level
of 0, 1 ppm, 2 ppm and 3 ppm. Observed parameters include the percentage of
life explants, the percentage of explants forming shoots, time of shoots, time of
roots, number of shoots, shoot height, number of roots and number of leaves. The
results showed that the administration of NAA and BAP had no significant effect
on the percentage of life explants, the percentage of explants forming shoots and
height of the shoots, but the administration of NAA had a significant effect on the
time of roots, the number of shoots, the number of roots and the number of leaves,
the administration of BAP significantly affected the time of the shoots, the number
of shoots, the number of roots and the number of leaves, the interaction between
the two treatments did not significantly affect all observation parameters.
