Potato (Solanum tuberosum L.) is a source of carbohydrates and processed food. Conventional propagation is slow and disease-prone, so tissue culture is a solution to produce superior seedlings. This study aims to determine the interaction of BAP and NAA concentrations in MS media on potato shoot multiplication. This research was conducted at the Tissue Culture Laboratory, Alifa Agricultural Research Centre (ALIFA-ARC), Medan, from November 2024 to January 2025. This study used a completely randomised design (CRD) with two treatment factors, namely the concentration of BAP (0 mg/l; 0,5 mg/l; 1 mg/l; 1,5 mg/l) and NAA (0 mg/l; 0,5 mg/l; 1 mg/l; 1,5 mg/l) with four replications. Parameters observed included the percentage of living explants, percentage of explants forming buds, number of buds, shoot height, number of roots, root length, number of leaves, and plant height. The results showed that the percentage of living explants was 100%, but only 10.93% managed to form shoots. The interaction of BAP and NAA treatment did not significantly affect the number of shoots and shoot height, but significantly affected the number of roots and root length. The treatment of BAP 0 mg/l and NAA 1 mg/l (B0N2) produced the highest number of roots (11,75 strands), while the highest root length (22,04 cm) was found in the treatment without the addition of BAP and NAA (B0N0). Single BAP treatment significantly affects the number of leaves and plant height, with 0 mg/l BAP treatment (B0) showing the best results.
