Isolation and Identification of Class of Flavonoids Compounds from the Leaves of Flamboyan (Delonix regia (Bojer Ex Hook.) Raf.)
Abstract
Flamboyan (Delonix regia (Bojer Ex Hook.) Raf.) is a plant that contains phytochemical compounds that have health benefits and are efficacious as medicinal ingredients. Isolation and identification of flavonoid compounds from flamboyant leaves have been carried out through a maceration extraction process using methanol solvent. The methanol extract was concentrated and then repeatedly extracted with ethyl acetate until it tested negative with FeCl3 5%. The concentrated ethyl acetate extract was dissolved in methanol and partitioned with n-hexane. The methanol layer was then evaporated until all the methanol had evaporated. The concentrated methanol extract was then analyzed by Thin Layer Chromatography (TLC) and separated using column chromatography with silica gel as the stationary phase and chloroform:ethyl acetate (90:10, 80:20, 70:30, 60:40, 50:50, 40:60,
30:70, 20:80, 10:90) v/v as the mobile phase. The seventh fraction was selected for further purification using preparative thin-layer chromatography with silica gel as the stationary phase and chloroform:ethyl acetate (30:70) v/v as the mobile phase. The pure compound obtained was a yellow amorphous solid weighing 15.4 mg with an Rf of 0.57 using chloroform:ethyl acetate (30:70) v/v as the eluent. Based on UV-Vis Spectroscopy analysis with methanol solvent, two absorption peaks were observed at wavelengths (λmax) of 315 and 265 nm. FT-IR spectroscopy showed the presence of O-H, C-H, C=O Ketone, C=C Aromatic, C-H sp3, C-O, C-O-C, C-H sp2 groups, and Proton Nuclear Magnetic Resonance (1H-NMR) spectroscopy revealed the presence of protons H-3, H-6, H-8, H-2’, H-3’, H-4’, H-5’, H-6’, and OCH3. Based on the analysis data, the isolated compound is a flavonoid compound of the flavone class
