The Effect of Honey Addition with Dimethyl Sulfoxide (DMSO) on Sperm Quality After Cryopreservation in Common Carp (Cyprinus carpio)
Abstract
Cryopreservation is a reproductive technology used to preserve gametes at ultra-low temperatures in order to maintain sperm viability over a long period. This study aimed to determine the effect of honey addition as a natural extender combined with Dimethyl Sulfoxide (DMSO) on the motility and viability of common carp (Cyprinus carpio) sperm after cryopreservation. The research was conducted in March–April 2025 at UPTD BBI Pinang Mancung and the Artificial Insemination Laboratory of North Sumatra Province. The experimental design used was a Completely Randomized Design (CRD) with five treatments of honey concentration (0%, 0.25%, 0.5%, 0.75%, and 1%) and three replications for each treatment. The results showed that the addition of honey extender at different concentrations had a significant effect on sperm motility and viability (p<0.05). The treatment with 0.75% honey concentration (P3) resulted in the highest motility of 67.66% and viability of 72%, which was significantly different from the other treatments. These results indicate that honey can be an effective natural extender alternative to support the success of sperm cryopreservation in common carp.
