Anti-Inflammatory Activity Test Of Chitosan Microcapsules Of Purple Leaf Extract (GRAPTOPHYLLUM PICTUM L. GRIFF) On Raw 264 Cells Induced With Lipopolysaccharide
Abstract
Background: Inflammation is the body's attempt to activate or destroy invading organisms, remove irritants and set the stage for tissue repair. Anti-inflammatory drugs that are commonly used are divided into two major groups, namely steroidal anti-inflammatory drugs and nonsteroidal anti-inflammatory drugs. However, both groups of drugs have serious side effects on their users. Because of the many side effects of anti-inflammatory drugs commonly used today, more and more anti-inflammatory drugs derived from plants are being developed.
Objective: The purpose of this study was to determine the anti-inflammatory activity of chitosan microcapsules of purple leaf extract (Graptophyllum pictum L. Griff) against RAW 264.7 cells induced with lipopolysaccharide (LPS).
Methods: The research method carried out is an experimental method with the stages of research, namely, collection of materials, sample management, making chitosan-extract microcapsules from samples. The anti-inflammatory activity of chitosan microcapsules of purple leaf extract (Graptophyllum pictum L. griff) on RAW 264 cells induced with lipopolysaccharide was tested.
Results: This study tested the cytotoxicity activity of samples against lipopolysaccharide-induced RAW 264.7 cells at various concentrations, namely 31.25 ppm; 62.5 ppm; 125 ppm; 250 ppm; and 500 ppm. The average viability of live cells from these concentrations were 107,83%; 94,11%; 67,86%; 56,26%; and 37,11%, respectively, indicating the higher the concentration of the sample the lower the level of viability of live cells.
Conclusion: Based on the research results obtained, the results obtained were that at concentrations of 31.25; 62.5; 125; 250; 500 ppm in the form of cell viability percentage or percentage of live cells induced by lipopolysaccharide, the results were 107.83%; 94.11%; 67.86%; 56.26%; 37.11%. Thus, it can be concluded that chitosan-purple leaf extract microencapsulation has anti-inflammatory activity on lipopolysaccharide-induced RAW 264.7 cells at concentrations of 31.25 and 62.5 ppm. Cell viability at these concentrations is more than 80%.
