    Abstract
    Background : Jeruju leaves (Acanthus ilicifolius) have long been used in traditional medicine, especially in coastal areas and mangrove ecosystems, because they are known to have various pharmacological activities. Jeruju leaves are known to contain various bioactive compounds, one of which is rutin which has high antioxidant activity. Identification and determination of rutin levels in plant extracts are important as a basis for quality control and development of phytopharmaceutical preparations. Objective: This study aims to identify and determine the levels of rutin compounds in jeruju leaf extract using the Thin Layer Chromatography (TLC) method combined with densitometry. Method : Jeruju leaf extract was analyzed using TLC with a stationary phase of silica gel GF254 and a mobile phase of a mixture of Ethyl Acetate-n Butanol- As formate (5:4:1) and combined with quantitative analysis by densitometry. Result : The results of the analysis showed a blue stain at a wavelength of 366 nm with an Rf value of 0.37 nm which was identified as rutin based on comparison with the standard. Measurement of rutin levels using densitometry produced levels of 5.6913 mgRE/g extract. Conclusion : Jeruju leaf extract is proven to contain significant amounts of rutin compounds and has the potential to be developed as a source of natural antioxidants.
