dc.description.abstractBackground: The analysis of andrographolide in the extract of sambiloto (Andrographis paniculata) is crucial for ensuring the quality of herbal products in accordance with the principles of Good Traditional Medicine Manufacturing Practices (CPOTB). The Thin Layer Chromatography-Densitometry (TLC- Densitometry) method was chosen for its ability to accurately identify and quantify andrographolide. This study aims to ensure the consistency and effectiveness of sambiloto products, thereby supporting the use of safe and high-quality natural remedies. Objective: This study aims to determine whether the compound andrographolide can be analyzed qualitatively and quantitatively using TLC-Densitometry and to measure the concentration of andrographolide in sambiloto leaf extract. Method: The method involves separation using silica gel plates as the stationary phase and a mixture of n-hexane and ethyl acetate as the mobile phase. Qualitative and quantitative analyses are performed using the TLC-Densitometry method. The procedure includes sample preparation, qualitative analysis through observation under UV light, and quantitative analysis to determine the concentration of andrographolide based on the intensity of the spots. Result: The qualitative analysis of andrographolide in sambiloto leaf extract (Andrographis paniculata) was conducted using Thin Layer Chromatography (TLC) with the aid of a UV Visualizer, identifying the presence of andrographolide. The results showed spots that corresponded to the standard andrographolide. The quantitative analysis revealed that the concentration of andrographolide in the sambiloto leaf extract was 8.8361 mgAE/g extract or 0.8%. Conclusion: The Thin Layer Chromatography (TLC) Densitometry method is effective in detecting the presence of andrographolide in sambiloto leaf extract and provides accurate information regarding the concentration of this compound. The andrographolide was clearly detected at an Rf value of 0.50, indicating the presence of andrographolide with a concentration of 8.8361 mg GAE/g extract.
