This study aimed to determine the effect of adding various types of milk
(cow’s milk, goat’s milk, and skim milk) to tris egg yolk extender on the quality of
Murrah buffalo (Bubalus bubalis) spermatozoa after the freezing process. The
research was conducted at the Artificial Insemination Center (UPTD IB) of North
Sumatra using a Completely Randomized Design (CRD) in a factorial pattern
with two factors: type of milk (3 types) and concentration (3%, 6%, and 9%), each
with 3 replications, resulting in 27 experimental units. Parameters observed
included motility, viability, and abnormality of spermatozoa after thawing. The
results showed that both the type of milk and its concentration had a highly
significant effect (P<0.01) on all three parameters. The use of 9% cow’s milk
yielded the best results, with motility at 34.2%, viability at 43.2%, and
abnormality at 15.3%, which are still within the normal distribution limits for
frozen semen. Conversely, the 9% skim milk treatment showed the lowest motility
and viability values and the highest abnormality compared to other treatments.
The nutritional content and fat composition of each milk type are believed to
influence spermatozoa quality after freezing. Therefore, the combination of 9%
cow’s milk in tris egg yolk extender is the most effective in maintaining the quality
of frozen Murrah buffalo semen.
