dc.description.abstractBlack cumin is a medicinal plant used in traditional medicine. Black cumin has numerous health benefits. Black cumin production in Indonesia is still low due to the country's less than optimal climate. However, the potential for black cumin production in Indonesia can be developed with appropriate cultivation techniques. Tissue culture is a vegetative cultivation technique that is feasible because it is not directly affected by climate change and can increase the content of secondary metabolites through the induction of polyploidy callus. This study aims to determine the increase in secondary metabolites in polyploidy callus of black cumin (Nigella sativa) plants through in vitro culture. This research was conducted at the Biotechnology Laboratory, Faculty of Agriculture, University of Sumatera Utara. from May to September 2025. This study used a Completely Randomized Design (RAL Method) with two factors. The first factor is the addition of colchicine concentrations: 100 ppm, 150 ppm, 200 ppm, and 250 ppm. The second factor is the soaking time: 4 hours and 6 hours. The results of this study indicate that increasing colchicine concentration and its interaction can increase the levels of secondary metabolites in black cumin (Nigella sativa) callus. Colchicine concentration also significantly affected the final callus weight, but soaking time had no significant effect on this parameter.en_US
