Soursop - Orange Boost : A Functional Drink Based on Soursop Leaf Extract (Annona muricata L.) and Siam Pontianak Orange Juice (Citrus nobilis) with the Addition of Stevia Sweetener as Cardiovascular Protection and Cancer Prevention
Abstract
This study was aimed to determine the comparison of soursop leaf extract with
Siam Pontianak orange juice and the addition of stevia sweetener, as well as its
potential in cardiovascular protection and cancer prevention in experimental mice.
The research employed a Completely Randomized Factorial Design (CRD) with two
factors: the ratio of soursop leaf extract to siam Pontianak orange juice (P1 = 70:30%,
P2 = 60:40%, and P3 = 50:50%) and the addition of stevia sweetener (S1 = 0.1%, S2 =
0.3%, and S3 = 0.5%).
The results showed that the ratio of soursop leaf extract and siam Pontianak
orange juice had a highly significant effect (P<0.01) on antioxidant activity (IC50),
vitamin C, total sugar, total acid, pH, color hedonic, aroma hedonic, taste hedonic,
and overall acceptance. The addition of stevia sweetener in the drink gave a highly
significant effect (P<0.01) on antioxidant activity (IC50), vitamin C, total sugar, total
acid, pH, color hedonic, taste hedonic, and overall acceptance, and showed a non-
significant effect (P>0.05) on aroma hedonic. The interaction between the ratio of
soursop leaf extract and siam Pontianak orange juice with the addition of stevia
sweetener had a highly significant effect (P<0.01) on antioxidant activity (IC50), pH,
and taste hedonic, and a non-significant effect (P>0.05) on vitamin C, total sugar, total
acid, color hedonic, aroma hedonic, and overall acceptance. The best treatment
determined using the De Garmo method was P3S2, namely 50% soursop leaf extract,
50% siam Pontianak orange juice, and 0.3% stevia sweetener. The Total Plate Count
(TPC) of P3S2 was 10.5 × 10 CFU/ml, meeting the requirements of SNI 3719:2022 fruit
juice beverages. Administration of the P3S2 sample at doses of 10,5 and 21 mg/g body
weight in mice reduced inflammatory cells and necrosis caused by oxidative stress in
the brain and heart induced by Benzo(a)pyrene.
