Introduction: Burns are a serious public health burden with a high incidence and mortality rate in Southeast Asia, particularly in Indonesia. Current burn management relies on wound dressings and ointments, but the efficacy of these interventions lacks supporting evidence. Previous studies indicate that the therapeutic properties of Peperomia pellucida potentially contribute to accelerating the healing of burn wounds. Objective: This study was undertaken to assess the healing effect of Peperomia pellucida nanoemulsion (PPNE) in a model of deep second-degree burn in Wistar rats. Materials & Methods: A deep second-degree burn was induced on the dorsal region of 36 male Wistar rats using a hot metal plate, and they were divided randomly into a negative control group (NC; NaCl 0.9%), a positive control group (PC; Moist Exposed Burn Ointment®), and a PPNE group. Wounds would be measured with a digital caliper on days 3, 5, 7, 9, 11, 13, and 14. Collected data would then be analyzed with one-way ANOVA test and Kruskal-Wallis’s test combined with post-hoc analysis Tukey’s HSD and Dunn’s Test. Results: The PC and PPNE groups showed acceleration of wound closure compared to NC group. Significant wound closure was found in the PC group on days 13 (p=0.044) and 14 (p=0.037) compared to the NC. PPNE exhibited diminished wound closure relative to the PC group, indicating its suboptimal wound-healing efficacy in burn cases. Conclusion: PPNE shows inferior burn wound-healing ability. Further studies are needed to explore its properties. Keywords: burn, Peperomia pellucida, moist exposed burn ointment®, wound healingen_US
