    Isolation and Identification of Flavonoid Compounds from Red Kamala Stem Bark (Mallotus Philippensis (Lam.) Müll.Arg)

    Abstract
    Isolation and identification of flavonoid compounds from the bark of red kamala (Mallotus Philippensis (Lam.) Müll.Arg) has been carried out through maceration extraction stages using methanol solvent. The methanol extract was concentrated and then dissolved with ethyl acetate. The ethyl acetate extract was evaporated and concentrated. The concentrated ethyl acetate extract was dissolved with methanol and partition extracted using n-hexane. The methanol layer was evaporated until all the methanol had evaporated. The concentrated methanol extract was analyzed by TLC, then separated by column chromatography with a stationary phase of silica gel and a mobile phase of n-hexane: ethyl acetate with a ratio of (90:10, 80:20, 70:30, 60:40, 50:50, 40:60, 30:70, 20:80, 10:90) v/v. A pure compound in the form of a yellowish white amorphous solid was obtained, as much as 6.3 mg with Rf = 0,70 using n-hexane: ethyl acetate (10:90) v/v as eluent. Based on UV-Vis spectrophotometer analysis with methanol solvent, it showed two absorption peaks with wavelengths (λmax) of 315 nm and 264 nm. The FT-IR spectrum showed the presence of O-H, C-H sp3, C=O ketone, C=C aromatic, C-H, C-O-C and C-H sp2 groups respectively at wave numbers 3388.93; 2927.94; 1724.36; 1514.12; 1379.10; 1276.88; 871.82 cm-1. The Proton Nuclear Magnetic Resonance (1H-NMR) spectrum showed the presence of protons H-6; H-8; H-3' and H-5' and H-2' and H-6'. Spectroscopic data indicate that the isolated compound is a flavonol.
