Black cumin is a type of seasonal plant as a medicinal plant that is often used in traditional medicine. To meet the needs of black cumin appropriately, it can be done through tissue culture. Tissue culture is a technique of vegetative plant propagation by isolating parts of the plant and growing it in a controlled artificial medium to be able to produce new individuals that are identical to the addition of the right growth regulator. This study aims to determine the effect of the use of NAA growth regulator and BAP growth regulator as well as the combination interaction of NAA and BAP on the growth of black cumin shoots in vitro. This research was carried out at the Biotechnology Laboratory, Faculty of Agriculture, University of North Sumatra from February 2025 to July 2025. This study used the Complete Random Design (RAL) method with 2 treatment factors and each treatment had 3 replicates. The first treatment factor is the concentration of ZPT NAA which consists of O ppm, 0.5 ppm, and 1 ppm. The second treatment factor is the concentration of ZPT BAP which consists of O ppm, 2 ppm, and 4 ppm. The parameters observed were the percentage of explants forming shoots (%), the percentage of explants forming calluses (%), the length of shoots, the time of bud emergence, and the time of leaf emergence. The results of this study show that the administration of BAP has a real effect on the observation of shoot length and the time of bud emergence. The administration of NAA had no significant effect on the parameters of bud length and the time of emergence of black cumin buds. The combination interaction of NAA and BAP had no significant effect on the parameters of bud length and bud emergence time.
